Enhanced Biosafety Level 2 (BSL-2+) Guidance
This page outlines general guidance to be followed when laboratory activities have been approved by the Boston University (BU) Institutional Biosafety Committee (IBC) for BSL-2+ containment. Additional requirements may apply based on the risk assessment of individual agent and procedures.
In general, BSL-2+ builds upon BSL-2 practices with the following minimal additions:
- Modified PPE.
- Double gloves.
- Disposable gown or Tyvek suit that is removed before leaving the containment space.
- Respiratory protection using an N95 or PAPR
- Sink for handwashing and eyewash within the containment space
- Ensure the lab has a negative airflow
In addition to these requirements investigators should consult their EHS representative for specific project needs. Investigators need to also ensure they are compliant for operations of a BSL-2+ laboratory, which at BU requires:
- Determine whether materials must be autoclaved prior to removal from the facility. Contaminated solid waste (pipettes, tips) is treated with freshly prepared household bleach solution to a final concentration of 10% for 30 minutes (or any other EPA approved disinfectant) before discarding in biohazard bag with a lid. Liquid waste is similarly treated with bleach solution to a final concentration of 10% for a minimum of 30 min prior to disposal in the sink.
- Lab-specific training emphasizing the specific BSL-3 practices is provided by the PI to all lab personnel.
- Lab personnel must participate if medical surveillance is required per direction of IBC and occupational health physician.
- In addition to the institution’s biosafety manual, a specific SOP is developed by the PI to detail appropriate BSL-3 practices for the project.
- All work with infectious virus or with infected cells is performed in a biosafety cabinet (BSC), including loading/unloading centrifuge rotors/cups.
- Disposable, solid-front, fluid-resistant gowns are practical. Additionally, double gloves, respirator, face, and eye protection is required, as determined by risk assessment.
