To meet increasing demand for tailored mass spectrometry applications across BU campuses, the TDPL has partnered with BUSM’s Proteomics Service Center (PSC) led by Dr. Joseph Zaia to strategically triage experiment requests toward “the best instrument for the job.” The PSC boasts two Thermo Scientific Orbitrap Exploris 480 mass spectrometers, established workhorses in proteomics workflows requiring high-resolution, accurate mass determinations with deep proteome coverage. The PSC offers both TMT-multiplexed and label-free proteomics workflows. The PSC also offers affinity proteomics of pulldowns including proximity labeling proteomics as well as on-tissue slide proteomics and glycomics and laser microdissection proteomics. The PSC is also a global leader in mass spectrometry-based glycomics and offers other post-translational modification (PTM) services including phosphoproteomics and glycoproteomics.
For more bespoke applications, the TDPL harbors a Thermo Scientific Orbitrap Eclipse Tribrid mass spectrometer, with several specialized capabilities: High Mass Range MSn (HMRn), Proton Transfer Charge Reduction (PTCR) and a multitude of dissociation techniques, including CID, HCD, ETD, EThcD and UVPD, providing a catalogue of options for intact protein analysis. The Eclipse also enables highly accurate quantitation of multiplexed (TMT) experiments using real-time database searches in combination with synchronous precursor selection and MS3-based quantitation (RTS-SPS-MS3), which is ideal for experiments requiring the highest standard of quantitative accuracy.
