And the problem is getting worse,<span>\u00a0<\/span><a href=\"https:\/\/www.cdc.gov\/drugresistance\/covid19.html\" target=\"_blank\" rel=\"noreferrer noopener\">fueled in part by the COVID-19 pandemic<\/a>: more pathogens are finding ways to sneak around our current antibiotics.<\/p>\n<p class=\"is-style-default\">But the fightback is on, and being led by a Boston University\u2013based nonprofit partnership that\u2019s spent the past eight years accelerating the development of new antimicrobials.<span>\u00a0<\/span><a rel=\"noreferrer noopener\" href=\"https:\/\/carb-x.org\/\" target=\"_blank\">CARB-X<\/a><span>\u00a0<\/span>directs funding and expertise to companies and researchers around the world to spur innovative antibiotics, vaccines, and rapid diagnostics. Many are happening in university labs or at small start-ups; CARB-X has supported developers in a dozen countries.<\/p>\n<p class=\"is-style-default\">\u201cWe didn\u2019t know it in 2016, but CARB-X has become one of the key global initiatives to save the future of antibiotics,\u201d says<span>\u00a0<\/span><a href=\"https:\/\/www.bu.edu\/law\/profile\/kevin-outterson\/\" target=\"_blank\" rel=\"noreferrer noopener\">Kevin Outterson<\/a>, CARB-X\u2019s executive director and Austin B. Fletcher Professor of Law at BU School of Law. \u201cThrough funding and an innovative support model for small and dedicated drug discovery teams, CARB-X has taken a leadership role in replenishing the early pipeline of potential new antibiotics, preventatives, and diagnostics for bacterial infections at a critical time when large pharmaceutical researchers and private investors have focused their energies elsewhere.<\/p>\n<p class=\"is-style-default\">\u201cIf CARB-X had not been around, we would have seen less innovation, fewer R&amp;D projects progressing toward patients, and a lower survival rate of dedicated research teams.\u201d<\/p>\n<p class=\"is-style-default\">CARB-X has also driven two diagnostic products to market,<span>\u00a0<\/span><a rel=\"noreferrer noopener\" href=\"https:\/\/www.bu.edu\/articles\/2019\/carb-x-superbug-detector\/\" target=\"_blank\">including a superbug detector<\/a><span>\u00a0<\/span>developed by Lexington, Mass.\u2013based T2 Biosystems. That device speeds up blood screening, giving doctors a crucial head start on spotting antibiotic-resistant organisms and building a treatment plan.<\/p>\n<p class=\"is-style-default\">\u201cBoth products seek to reduce the time between when a patient presents with sepsis symptoms and when a proper antibiotic is administered,\u201d says<span>\u00a0<\/span><a href=\"https:\/\/carb-x.org\/team\/erin-duffy-phd\/\" target=\"_blank\" rel=\"noreferrer noopener\">Erin Duffy<\/a>, CARB-X\u2019s chief of research and development. \u201cThis is important, because the likelihood of death<span>\u00a0<\/span><a rel=\"noreferrer noopener\" href=\"https:\/\/www.atsjournals.org\/doi\/10.1164\/rccm.201609-1848OC\" target=\"_blank\">increases by nearly 10 percent<\/a><span>\u00a0<\/span>for every hour in that time gap.\u201d<\/p>\n<p class=\"is-style-default\">CARB-X is funded by a consortium of governments and foundations that includes the US, German, UK, and Canadian governments, the global charitable trust Wellcome, the Bill &amp; Melinda Gates Foundation, and the Novo Nordisk Foundation. The institute is developing compounds that show early promise against pathogens that cause a type of lower respiratory tract infection known as community-acquired bacterial pneumonia.<\/p>\n<p class=\"is-style-default\">\u201cWith this 100th project, we are doubling down on our support of novel approaches to deliver antibiotics that clinicians and patients need,\u201d says Duffy. \u201cIf successful, the HIPS project will offer a workhorse antibiotic for community-acquired infections that will also take the pressure off antibiotics in the [World Health Organization] model list of essential medicines.\u201d<\/p>\n<p class=\"is-style-default\">According to Duffy, CARB-X will continue to focus on the bacteria and infections that \u201cdrive the greatest burden of disease\u201d and products that have the potential to save millions of lives. The nonprofit partnership<span>\u00a0<\/span><a href=\"https:\/\/carb-x.org\/carb-x-news\/2024-funding-round\/\" target=\"_blank\" rel=\"noreferrer noopener\">just announced a new global funding round<\/a><span>\u00a0<\/span>and is calling for project proposals in four areas: therapeutics for infections caused by gram-negative pathogens, novel approaches to the prevention of invasive disease, diagnostics for neonatal sepsis, and diagnostics for diagnosing lower respiratory tract infections.<\/p>\n<p class=\"is-style-default\">\u201c<a href=\"https:\/\/www.thelancet.com\/journals\/lancet\/article\/PIIS0140-6736(22)02185-7\/fulltext\" target=\"_blank\" rel=\"noreferrer noopener\">Bacteria kill 7.7 million people<\/a><span>\u00a0<\/span>a year. A growing percentage of those people die from a drug-resistant bacterial infection, already killing more people each year than HIV\/AIDS and malaria combined,\u201d says Outterson. \u201cEvolution pushes bacteria to resist antibiotics, so this problem only gets worse unless we use our brains and pocketbooks.
