Abstract: This prospectus consists of two different projects to obtain volumetric information in a single acquisition. The development of high resolution, 3D imaging endomicroscopy has recently been gaining interest due to needs in in vivo studies of neuronal dynamics and interactions in deep brain tissue. However, most types of endomicroscopy techniques adopting micro-objectives can hardly perform 3D imaging due to the lack of minimally invasive scanning mechanism. We present a GRIN-lens based fluorescence endomicroscopy capable of obtaining pseudo 3D image of a fluorescent sample without scanning the probe. Several approaches are being developed for fast, large-scale volumetric imaging due to the growing interest in the study of neural networks to understand the brain dynamics leading to sensory experiences and behavior. We present a novel microscopy technique, capable of ultrafast, multiplane, optically sectioned imaging over large fields of view on the millimeter scale for in vivo calcium and voltage imaging.
