Introduction to the Flow Cytometry Core Facility

Welcome to the BUMC Flow Cytometry Core Facility. We provide flow cytometry services for analysis and sorting of cells, as well as expert consultation for project/experiment planning. We have three FACScan analyzers, one FACSCalibur analyzer, one LSRII analyzer, and one MoFlo cell sorter.

LSRII
  • Location: 670 Albany St., BioSquare III building, 5th floor
  • Rates: BUMC: Self-$45.00/hr; Assisted-$67.50/hr; non-BUMC: 150% of BUMC
  • Description: The LSRII is a four laser, 12 detector analysis flow cytometer capable of detecting 10 colors simultaneously. We have a "Blue" (488nm) laser, a "UV" (355nm) laser, a "Red" (633nm) laser, and a "Violet" (405) laser. This unit also features a high throughput 96 well autosampler. Digital acquisition increases sampling rates compared to analog machines. The acquisition workstation is a PC running FACS Diva 6.0. The data files can be batch converted for analysis using other applications such as Summit, CellQuest Pro, and WinMDI to name a few.

FACSCalibur

  • Rate: BUMC: Self-$40.00/hr; Assisted-$60.00/hr; non-BUMC: 150% of BUMC
  • Description: The FCCF FACSCalibur is a single laser, five detector analysis flow cytometer. The laser excites at 488nm and collects data for forward scatter, side scatter, Fl1 (530±30nm), Fl2 (585±42nm), and Fl3 (650±30nm). There is a Mac computer running the FACSCalibur using BD Cellquest Pro v5.2 software.

L508A FACScan

  • Description: The L901 FACScan is a single laser, five detector analysis flow cytometer. The laser excites at 488nm and collects data for forward scatter, side scatter, Fl1 (530±30nm), Fl2 (585±42nm), and Fl3 (650±30nm), thus making it a three color analyzer. There is a MacG4 running the FACScan using BD Cellquest Pro v5.2 software. A MacG4, also running BD Cellquest Pro v5.2 software, serves as an analysis workstation and is not connected to the FACScan.

X620 FACScan

  • Description: The X620 FACScan is a single laser, five detector analysis flow cytometer. The laser excites at 488nm and collects data for forward scatter, side scatter, Fl1 (530±30nm), Fl2 (585±42nm), and Fl3 (650±30nm), thus making it a three color analyzer. There is a MacG5 running the FACScan using BD Cellquest Pro v5.2 software. A MacG4, also running BD Cellquest Pro v5.2 software, serves as an analysis workstation and is not connected to the FACScan.

MoFlo High Speed Cell Sorter
  • Location: 670 Albany St., BioSquare III building, Rm 527
  • Rates: BUMC: Assisted only - $80.00/hr; non-BUMC: 150% of BUMC
  • Description: The MoFlo is a three laser, seven detector flow cytometer capable of sorting cells into two populations based on user specifications/gating. The MoFlo has one 488nm laser, a UV laser tuned to 351nm, and a red laser at 635nm. Like the FCCF FACScans, the MoFlo collects data for forward scatter, side scatter, Fl1 (530±30nm), Fl2 (585±42nm), and Fl3 (650±30nm) but also has detectors for four more channels which can be adjusted through the use of filters. MoFlo is capable of sorting cells into two distinct populations, either into tubes or into microtiter plates of any size from 6 to 96 wells. Single cell deposition is also possible.
